Fatty Acid Profiling – Food Oils, Marine Oil, Vegetable Oils, Nutraceutical Oils

The quantity and quality of the fatty acids play an essential part in human diet and health. Imbalances of fatty acids can significantly influence inflammatory disorders, cardiovascular disease, pregnancy, hyperactivity, depression and many other conditions. Edible oils consist mainly of fatty acids (>90%) which are structurally differentiated by their chain length, cis/trans orientation, the position of their double-bond(s), and their ratio of saturated and unsaturated fatty acids. GC analysis provides a comprehensive fatty acid profile that allows practitioners to accurately assess dietary intake and metabolism of fatty acids.

Analytical Targets: Fatty acid methyl esters including but not limited to - Eicosapentaenoic Acid (EPA), Docosahexaenoic acid (DHA), Omega-3 Fatty Acids (C18:3n3, C20:3n3, C20:5n3, C22:5n3, C22;6n3), Omega-6 Fatty Acids (C18:2n6, C18:3n6, C20:2n6, C20:3n6, C20:4n6, C22:2n6, C22:4n6) amongst others.

Method Summary

Omega 3 Oils

This method determines the fatty acid composition of marine oils in relative (area %) values, and EPA and DHA in absolute (mg/g) values using a bonded polyglycol liquid phase in a flexible fused silica capillary column. The esterification of these fatty acids to fatty acid methyl esters is performed using an alkylation derivatization reagent. Methyl esters offer excellent stability and provide quick and quantitative samples for GC analysis. GC analysis provides a comprehensive profile that allows practitioners to accurately assess the content and profile of fatty acids in their oil product.

Fatty acids are extracted from samples according to the method of Bligh & Dyer (1) in the presence of the internal standard. Subsequent processing using methods similar to those described in (1). The fatty acid methyl esters are transmethylated. The resulting fatty acid methyl esters are then analyzed on an Agilent 7890B gas-liquid chromatograph with a 60-m DB-23 capillary column (0.32 mm internal diameter) against their internal standard. Data is reported in ug/g and % by weight.
